As well during high pressure as during low pressure gradient forming, solvents are generated that are firstly only mixed together but not completely stirred. During the low pressure gradient forming the pumps suck the different components in segments and forward them to the injection valve. If there is no mixing chamber between pump outlet and injector inlet, the UV detectors will have a noisy basis line and the retention times will be irregular. |

Insufficiently degassed mobile phase in HPLC will lead to higher baseline noise, spikes due to air bubbles, and significant drifting and oscillation of the detector signal in HPLC. These phenomena may lead to significant difficulties in integration of peaks and create problems for quantitative analyses. Dissolved gases affect the operation of the pump and may cause strong pulsation and flow rate changes. This results in serious retention time shifts, making analyte identification difficult. |

When Micro HPLC or Nano HPLC is employed to separate samples, the components of the
sample are isolated as fractions in the same way as when analytical scale HPLC is used. In many
laboratories, the fractions are deposited into microtiter plate wells (MTP) or are spotted onto
MALDI-MS targets for further investigation. The SunCollect MALDI-Spotter is extremely successful in collecting fractions using either approach. If desired, a single fraction can be saved in both formats at the same time. |
